The Den of Evil is the very first dungeon in the game, located in the Blood Moor. It is the subject of the first quest, where one is tasked with entering the Den of Evil and killing all the monsters there. Completing the quest rewards the player with an extra skill point. The most notable inhabitants of the cave is probably the first super unique monster met by the player, the Corrupted Necromancer.
